Rat liver nuclei were purified and incubated with isolated viral capsids in the presence of cytosol, fixed, and processed for electron microscopy. In ultrathin plastic sections, HSV-1 capsids that bound to NPCs in the nuclear membrane were easily recognized by their size (125 nm) and their sixfold symmetry. The viral DNA is still visible as the electron-dense core inside the capsids. The capsids seem to interact with cytoplasmic fibrils emanating from the cytoplasmic face of the NPCs.
