All Rights Reserved.): hFbh1, the human counterpart of Saccharomyces cerevisiae Srs2 DNA helicase, other than acting as helicase, suppresses the requirement for postreplication repair (PRR) in yeast via its F-box domain and the interaction with the Skp\NCullin\NF-box (SCF) machinery. We propose that the two-domain structure allows hFbh1 to act as a Janus Geminus in facing the requirement of homologous recombination and PRR in front of a lesion. Shown is a possible structure for the Fbh1-SCF complex, obtained by juxtaposing the structures of UvrD/DNA (light blue) and SCF (pink; Skp2 in blue) overlapped with a statue representing Janus.
